50 Cent se narodil jako Curtis Jackson, 6. července 1976 v South Jamaica / Queens v New Yorku, do rodiny s drogovou minulostí. Pseudonym 50 Cent si Curtis zvolil podle proslaveného gangstera Fort Green Projects. Rapper je věřící a jedno z jeho tetování říká: „Bůh mi rozumí.“ Otce nikdy nepoznal a matka byla dealerkou drog, vystupující pod přezdívkou Fat Cat. Fifty u něj podepsal svou první smlouvu. Jam Master Jay ho naučil sestavovat písně a dodržovat správný rytmus.Díky tomu se Fiftyho mixtapes začali slušně prodávat. I přestože se od Jam Master Jaye naučil spoustu věcí, brzy pochopil, že se s ním nedostane tam kam potřebuje. Smlouvu vypověděl a začal se poohlížet po větší nahrávací společnosti. V roce 1998 50 Cent, Lloyd Banks a Tony Yayo zakládají G- Unit.
V roce 1999 zaujal 50 Cent producentské duo Trackmasters, proslavené několika platinovými alby např. J. Lo - Jennie From The Block nebo All Stars Space Jam, a to ho představilo u Columbia Records. Fifty podepsal u Columbia Records smlouvu a hned začal pracovat na svém debutovém albu Power Of The Dollar. Při nahrávání tracků strávil 2 a půl týdne zavřený ve studiu, o čemž se s nelibostí zmiňuje ve své autobiografii. Album Power Of A Dollar nebylo oficiálně nikdy vydáno, došlo jen k jeho propagaci. Některé časopisy, např. Blaze, označili Fiftyho debutový počin za klasiku. O pozdvižení se postarala skladba How To Rob An Industry Nigga, kde se Fifty představil jako hladový, šílený nováček, který touží po okradení známých rapperů. Mezi „napadenými“ nechyběli producenti, rappeři a r’n’b zpěváci zvučných jmen jako: Jay-z, Busta Rhymes, Rza, Raekwon, Timbaland, Big Punisher, Sticky Fingaz, ODB, Trackmasters, Ghostface Killah, Will Smith, Bobby Brown a Whitney Houston, Lil Kim, Mariah Carey, Missy Elliott, P. Diddy, DMX, Naughty By Nature a další. Ohlas byl obrovský, někteří samozřejmě odpověděli na povedenou parodii diss tracky. Jay-Z odpověděl dissem Go Against Jigga Your Ass Is Dense / I Am About A Dollar What The Fuck Is 50 Cent. Fiftyho odpověď Be A Gentleman mnoho lidí neslyšelo, protože u Columbia Records už nestačila vyjít. Některým kritizovaným se tento druh humoru buď líbí nebo zapomněli na hrdost, aby si později mohli ukrojit kousek z koláče slávy 50 centa. Tak třeba Missy Eliott si ho přizvala na své album Under Construction a Lil Kim ho i přes diskriminující slova featuje na soundtracku k 8 Mile v písničce Magic Stick. O pár let později Fifty o kontroverzním tracku How To Rob prohlásil: „Ta píseň byla nejlepší co se mi mohlo přihodit, nahrál jsme ji, protože jsem se potřeboval odlišit od spousty dalších rapperů na mém labelu. A povedlo se, najednou znal 50 Centa každý. Text jsem napsal cestou do studia, obsahoval pravdu a všichni to věděli. Krádež byla z důvodů mé tehdejší finanční situace pro mě aktuální téma. Ale byl to pochopitelně hlavně pokus o upoutání pozornosti.“
Půl roku po dokončení svého prvního alba, 24.května 2000, byl 50 postřelen před domem
svých prarodičů. Zasáhlo ho devět kulek. Fifty strávil v nemocnici sice jen 13 dní, ale stálo ho to smlouvu, protože Columbia Records se zalekla negativní publicity a rozšiřování nelegálních kopií Power Of The Dollar po internetu. Od tohoto incidentu se Fifty pohybuje téměř neustále s neprůstřelnou vestou a nechal dokonce vyrobit speciální vestu i pro svého syna. 50 se vyjádřil k útočníkovi v časopise Rolling Stone: „Syn smrti, kdo mě postřelil.“
Fifty se musel vrátit zpátky na ulici a pokračovat v dealování drog, aby měl peníze na mixtapes.V tomto období také vzniká rozvleklý beef s Ja Rulem. Ja Rule sleduje Fiftyho jak se v jednom klubu baví s týpkem, který mu před týdnem ukradl šperky, potom dojde před klubem k rvačce a několik členů Murder Inc. zraní Fiftyho nožem. 50 Cent spolu s Sha Money XL nahrál přes 30 songů a na jaře 2001 zveřejnil neoficiální LP Guess Who’s Back, které vzniklo přepracováním materiálu od umělců jako jsou Jay-Z nebo Raphael Saadiq. Protože stejný názor měl i Dr. Dre podepsal Fifty zanedlouho smlouvu u Shady/Aftermath Records. V té době také vrcholí beef s Ja Rulem. Jestli má spor co dočinění s vraždou Fiftyho mentora Jam Master Jaye v listopadu 2002 je ve hvězdách. Intenzivního vyšetřování se vraždě dostalo i z důvodu údajné souvislosti s drogami. Label Murder Inc. je údajně financován penězi jistého mafiánského bosse, se kterým měl Fifty shodou náhod problémy už za dob kdy se věnoval dealování drog. Ja Rule vedl proti Fiftymu očerňující kampaň, mimo jiné ho označil za policejního informátora. V roce 2002 Fifty spolu s G-Unitem vydává další dvě neoficiální LP pod názvem 50 Cent Is The Future a No Mercy, No Fear, kde se objevil i track Wanksta, který nebyl původně určen do rádií, ale stal se nejžádanějším hitem právě na rádiích v New Yorku a objevil se i na soundtracku k Eminemově filmovému debutu 8 Mile.
V roce 2003 vychází Fiftymu první oficiální album Get Rich Or Die Trying, kterého se první týden prodalo 872.000 ks, čímž překonalo i debut Snoop Dogga. Dr. Dre a Eminem umožnili 50 založení labelu G-Unit Records. U nového labelu podepsali původní členové G-Unitu Lloyd Banks, Tony Yayo a Young Buck a v roce 2003 vychází i společné album G-Unitu Beg For Mercy, po němž následují sólové projekty jednotlivých členů. Fifty také vydal DVD The New Breed, investoval do své vlastní oděvní firmy a stejně jako Jay-Z přivádí na trh vlastní kolekci bot ve spolupráci s firmou Reebok. I to je důkazem, že někdejší beef mezi nimi je minulostí. Proti spolupráci 50 s Reebokem se postavil konzervativní moderátor Bill O’Reilly. Jeho bojkot, ale neměl na prodej G-Unit sneakers nakonec žádný negativní dopad. Reklama vztahující se k tomuto produktu byla ve Velké Británii zakázána, z důvodu že obsahuje jeden z Fiftyho songů údajně propagujících násilí. V roce 2004 z iniciativy šéfa Interscope podepisuje u G- Unit Records The Game. 50 vydává další neoficiální LP Fiddy The Hottest Nigga.
V roce 2005 vychází Fiftymu album The Massacre, o rozruch se postaral track Piggy Bank, který je vyhlášením války některým rapperům jako jsou Nas, Jadakiss a Fat Joe. Důvodem k tomu jsou mimo jiné také jejich vazby s Ja Rulem. U labelu G-Unit podepisují Olivia, DJ Whoo Kid, rapper Spider Loc a velkým překvapením jsou další přírůstky Mobb Deep, M.O.P. a Mase. Stále častěji se také objevují zprávy o neshodách nebo dokonce beefu mezi 50 Centem a Gamem. Game byl dokonce vyhozen z labelu G-Unit Records, o něco později ho ale Fifty vzal zpátky a veřejně se usmířili na tiskové konferenci, při příležitosti založení charitativní organizace G-Unity. I přes velký úspěch jejich duetů Hate It Or Love It a How We Do mají spolu spíš špatný vztah. Fifty se v roce 2005 objevuje i jako hlavní postava hry Bulletproof a znovu je mu vyčítáno, že propaguje násilí. Na názor, že tato hra může někoho poškozovat Fifty reaguje: „ Člověk, který se nechá ovlivnit počítačovou hrou musí podle mě už nějak poškozený být. Takovou osobu by v podstatě muselo negativně ovlivňovat téměř všechno.“ 50 Cent vydává také svou autobiografii v knižní podobě pod názvem From Pieces To Weight : Once upon a time in Soutside Queens. Pod názvem Get Rich Or Die Trying vychází 50 filmový debut, inspirovaný jeho životem a soundtrack k tomuto filmu.
